چکیده
single Sep-Pak has been used for as many as 10 serum samples with no noticeable loss in efficiency. For analysis of clinical samples, I compared the above sample pretreatment procedure to the acetonitrile extraction of Kabra et al. (1). The chromatographic conditions were similar to those of Kabra et al. (1), except for our use of ambient temperature and a 3.5 mL/min flow rate. The Sep-Pak pretreatment procedure resulted in samples that were significantly “cleaner” than the acetonitnile extracts when chromatographed. The height and duration of the extraneous peaks eluting near the solvent front were greatly reduced and the baseline was re-established before the anticonvulsants emerged. Hexobarbital can be used as an internal standard for this procedure. This sample pretreatment procedure allows for reproducibility and sensitivity with less column degradation.

Development and validation of a liquid-chromatographic procedure for serum creatinine.
We evaluated and compared three analytical procedures for measuring creatinine by testing pooled sera and sera from individual human subjects. In two of the methods, an enzymic and a conventional colorimetric procedure, creatinine was separated from serum proteins with tungstic acid and detected at 520 nm after reaction with alkaline picrate (Jaff#{233} reagent). The amount of enzymaticaIly act...
متن کاملMetabolism of hyperpolarized [1‐13C]pyruvate through alternate pathways in rat liver
The source of hyperpolarized (HP) [(13)C]bicarbonate in the liver during metabolism of HP [1-(13)C]pyruvate is uncertain and likely changes with physiology. Multiple processes including decarboxylation through pyruvate dehydrogenase or pyruvate carboxylase followed by subsequent decarboxylation via phosphoenolpyruvate carboxykinase (gluconeogenesis) could play a role. Here we tested which metab...
متن کاملEnzymic urea assay: a new colorimetric method based on hydrogen peroxide measurement.
We describe a new enzymic colorimetric method in which urea is measured in serum by use of a single reagent mixture. Ammonia produced by urea hydrolysis, catalyzed by urease, reacts with glutamate and ATP in the presence of glutamine synthetase. The ADP so produced is assayed in reactions catalyzed sequentially by pyruvate kinase and pyruvate oxidase in a system that generates hydrogen peroxide...
متن کاملalpha-Amylase activity by the Beckman reaction system and suppression by pyruvate.
We evaluated the new Beckman Enzymatic Amyalse-DS Method with maltotetraose as substrate. Our findings indicate that the method is approximately twice as sensitive as the old method involving soluble starch [Clin. Chem. 24, 762 (1978)]. The new method also shows a linear rate of reaction, in contrast to the curvilinear rate previously observed with the old method. The Km with maltotetraose is 0...
متن کاملImproved method for determination of high-density-lipoprotein cholesterol II. Enzymic determination of cholesterol in high-density lipoprotein fractions with a sensitive reagent.
A reagent is described for the colorimetric enzymic determination of high-density-lipoprotein (HDL) cholesterol. The reagent can be used with HDL fractions isolated by the various methods of precipitation of low- and very-low-density lipoproteins we investigated. The considerable sensitivity obtained by use of Barham-Trinder's reaction allows the sample/reagent volume ratio to be decreased to 1...
